The reduction of cytochrome c by milk xanthine oxidase.

چکیده
The reduction of cytochrome c by xanthine oxidase and the competitive inhibition of this process by carbonic anhydrase and by myoglobin have been studied by kinetic and by equilibrium binding methods. Carbonic anhydrases isolated from bovine and from human erythrocytes differed strikingly in their ability to inhibit competitively the reduction of cytochrome c. The KS for cytochrome c was a function of the concentration of xanthine oxidase, as were K; for carbonic anhydrase and Ki for myoglobin, whereas K, for xanthine was invariant under the same conditions. Binding studies performed by a variety of methods indicated that carbonic anhydrase does not bind to xanthine oxidase. Carbonic anhydrase was found to be a potent inhibitor of the sulfiteoxygen chain reaction initiated either by the reduction of oxygen at an electrode or by xanthine oxidase plus xanthine. The data are consistent with the conclusion that xanthine oxidase, when catalyzing the aerobic oxidation of xanthine, generates an unstable reduced form of oxygen, presumably the superoxide anion, and that this radical is the agent which directly reduces cytochrome c and initiates the sulfiteoxygen chain reaction. Carbonic anhydrase and myoglobin appear to inhibit the reduction of cytochrome c and the initiation of sulfite oxidation by reducing the steady state concentration of the superoxide anion. It is proposed that they accomplish this end by catalyzing the following dismutation reaction:

Competitive Inhibition by Myoglobin of the Reduction of Cytochrome c by Xanthine Oxidase*
During a study of the reduction of cytochrome c by milk xanthine oxidase, it was observed that relatively impure preparations of cytochrome c were reduced more slowly than highly purified preparations. This difference was found to relate to the presence, in impure preparations, of a potent inhibitor of the reduction of cytochrome c by xanthine oxidase. This inhibitor was of interest because its...

متن کاملXanthine oxidase. V. Differential inhibition of the reduction of various electron acceptors.
Milk xanthine oxidase can catalyze the reduction of oxygen, cytochrome c, nitrate, ferricyanide, and various quinones and dyes by many aldehydes and purines. When acting upon its substrates, this enzyme can also initiate the aerobic oxidation of sulfite (1, 2) and induce the chemiluminescence of lucigenin and of luminol (3, 4). The reduction of cytochrome c by milk xanthine oxidase has been the...
متن کاملEffects of flavonoids on xanthine oxidation as well as on cytochrome c reduction by milk xanthine oxidase.
That flavonoids inhibit xanthine oxidase from cow milk was confirmed by measuring oxygen consumption with an oxygen electrode. In contrast, flavonoids did not inhibit glucose oxidase, another oxygen consuming enzyme. Among the flavonoids tested, quercetin, kaempferol, myricetin, chrysin, quercitrin, and morin were potent inhibitors of xanthine oxidase; their inhibition rates (%) were 80, 70, 69...
متن کاملOnion, a Potent Inhibitor of Xantine Oxidase
Onion (Allium Cepa) contains high levels of flavonoids. Although there are many studies indicating the inhibitory effects of flavonoids on xanthine oxidase, there is no report on the effect of onion on this enzyme. Therefore, in the present study, the inhibitory effects of onion on xanthine oxidase are investigated. Fresh filtered juice of onion was prepared and its inhibitory effect on guine...
